I want to do an experiment with soil, and want to take samples at a certain depth in 1 m2. In 1 m2 fit 25 20x20cm2 areas, is that true?

Correct.

1 m² can also be stated as 10,000 cm².

A 20 cm by 20 cm area is 400 cm², 25 of these would be 10,000 cm².
